Method for the in-situ detection of proteins in tissues using a specific antibody coupled to a chromogenic enzyme complex. The staining allows for a rough estimation of the expression level and the intracellular location of the target protein.
Many parameters substantially influence IHC, for example:
- Time to tissue fixation
- Time of tissue fixation
- Type of fixation
- Antigen retrieval/protocol optimization
- Section age
- Section thickness
